This page contains instructions and an example for submitting data to the Molecular Ecology Resources Primer Database. Please review each of the data fields and determine how you are going to fill each field before you begin. That way, if you have any questions, you may contact the administrator prior to being forced to abandon an entry halfway through.
A complete database submission consists of all species × primer combinations for which information has been generated. The simplest ways this may be accomplished is to enter in the data for species from which the marker was originally developed, submit this data to the database, and then either go "back" to the submission form by using the browser's back button or equivalent keystroke or use the "template" link on the record detail page. This way, the form should still be populated with your record's data, so that the amount of data entry is minimized and only certain fields will need to be changed.
| Species |
This field is for entering in the name of the species for which information is being reported. Please use the full binomial (e.g., Capsicum annuum) for each entry, without the authority, subspecies, or population information. If this information needs to be included, it may be included as a note appended to the species field (see species note below). |

Species notes could refer to specific populations, if population structure is important for understanding the applicability of the information presented. General instructions for notes: This field, and all others like it, are for adding in a number which will be used as a superscript to relate a note for a field to the text of the note. The text for all notes for an entry is entered into one field below. Notes should be used sparingly to add in information which will be generally important to researchers in the field. The numbers in note fields should be sequential down the form, 1N, as they are used.

| Locus Name |
This field is for the locus name. Please limit names to Roman alphanumeric characters (az, AZ, 09, and hyphens). If possible, please try to make locus names as unique as possible (you may search the database for locus names you are considering to see if there is anything similar already). |

| Marker Type |
Please select a marker type from the drop-down list. Single nucleotide polymorphisms (SNP) includes all cloned genes where sequence comparisons are made, either by sequencing or allele-specific amplification. Cleaved (or cut) amplified polymorphic sequences (CAPS) are PCR products that have been restricted to reveal polymorphism; the enzyme used should be indicated in the marker type note field. This list may be expanded in the future to accomodate new marker types. |

| Repeat Motif |
This field is for describing the repeat motif for cloned microsatellite alleles, and may be ignored for other types of markers or other species for which only amplification information is known. When filling out this field, you should indicate the number of repeats by subscripting, which is accomplished by bracketing the number or symbol to be subscripted with the html subscript tags: |

| Genome |
Select the genomic compartment of the marker from the drop-down list. Ribosomal DNA is allowed as a special subset of markers. |

| Database Accession ID |
Enter in the database ID of the original cloning record. |
| Database ID Note |
Enter in the database name where the accession was submitted. |
| Is this the species the locus was originally cloned from/described in? |
This field is for indicating whether this record is from the species from which the locus was originally derived. If so, enter a Y in the field; if not, enter in the full name of the species from which the locus was derived. For example, if I were reporting amplification of a microsatellite in Capsicum chinense that was oringinally cloned from Capscium annuum, I would enter Capsicum annuum in the field. The utility of this field presumes that the record for the original species also is/will be in the database. |

This field is for entering in the forward primer for the locus. Please enter the sequence in capitals in the 5'3' direction. |

| Annealing Temperature |
This field is for entering in the PCR annealing temperature used during amplification. A °C will automatically be attached. |

This field is for attaching information about the annealing temperature, which may include details about cycling conditions related to annealing temperature. |
| Number of Alleles |
This field describes the number of alleles observed in assays of the species or group described by this record. If amplification was attempted for group, but was unsuccessful, enter a 0 in this box and the record will read, "tried, but no amplification." |

This field may be used to attach information to the number of alleles, and could be used to relay information about the presence of null alleles or the size of population tested. General instructions for notes. |
| Size Range |
This field provides the size range of the detected alleles. Range may be indicated by either a hyphen or the html code for an en dash (–) as in the example. The latter provides this output: 155165. |

| Expected Heterozygosity |
This field is for entering in the expected heterozygosity if the size of population screened was adequate to calculate He. |
| He note |
This field may be used to attach information to the He measure. General instructions for notes. |

This field is for entering in the observed heterozygosity if the size of population screened was adequate to measure Ho. |
| Ho note |
This field may be used to attach information to the Ho measure. General instructions for notes. |
| Contact Name |
This field is for the contact authors name. |
| Contact Email |
This field is for the contact authors email. If no email address is available please enter a fax number by typing in "fax: number". |
| Notes |
This area is for entering in the text for the note fields given numbers above. This may be accomplished most easily by writing the note in a text processor, and then pasting the information in this field. The text for each note should have the number, a period, and then the note text, followed by the characters <p> which are the equivalent of a line break. If no notes are needed for an entry, please be sure to delete the text in this field before submitting the record to the database. |
